The first one sounds more natural, however they are both correct. Though instead of saying 'in a week', it would be 'next week'.
1. "I have a day off next week." The word 'a' can also mean 'one'.
For example;
"I have an apple." or "I have a book."
2. "I have one day off next week."
It's mostly down to preference, but people tend to say "a day" more than "one day" when talking about time off.

Both are okay, you could also say “I have the day off” if you already mentioned what day, I prefer the first one though!

The correct way would be "I only have a day off."
As before, you can also say "I only have one day off." Though I've heard the first one more.
