Both sentences will primarily be interpreted as asking how long they studied Japanese at a point in the past. The last one is more specifically asking how many hours they have spent on studying Japanese. However, the meaning is rather vague. It could be "how many hours have you spent on Japanese learning until now?" It could be "How many hours a day did you study Japanese when you were in university?"
On the other hand, the first one could be about their overall experience on Japanese learning. The possible answers would be "I majored Japanese for three years in university", "I started going for Japanese five years ago and passed N2 last year", and such.
