First one is better, but I'd phrase it a bit differently. Should is unnecessary here, so it would sound more like, "First walk to the house with the red roof, then turn left."

The difference between "the" and "a" in this scenario is specificity. If there's more than one house with a red roof, then you would use "a," but since this is directions and you want to be as specific as possible, hopefully from your sentence the listener can learn that there is only ONE house with a red roof. If there is more than one, you would need to specify "the second house with the red roof" or "the house with the red roof and the green door." Hope that clears it up!

@deadwate I see! I kept reading your answer to understand it. Yeah, there was only one red-roofed house up ahead. We could see that from a distance. So, in that case, is it okay to say “the house with a red roof”? 🙂
