Do I use the statement only when someone was introducing somebody to me?
Could I use the statement when it's first meeting and I and he is only here(only two people and it's not being introduced by someone).

You could use it, bit it seems a bit old fashioned to Americans. It's more common to say "it's nice to meet you."
