They mean pretty much the same thing.
The only nuance is that using "so much" adds emphasis, so it gives more importance to how very much they have to do. Either it is a very large amount of things they need to do. Or they may be feeling overwhelmed by how much there is to do. Or they are trying to make a point, to emphasize that they really are too busy to do you a favor so stop asking. But for some reason, they are emphasizing just how very much they have to do. They are not just busy, they are VERY busy.
So they can mean the same thing, and be used in the same way. But sometimes people will use "I have so much to do" to extra emphasize just how extra busy they are.

They’re very similar, but “so much” is a little stronger than “a lot”.
In this sentence, “so much” sounds like you are unhappy/overwhelmed by the amount of work and you’re complaining/venting about it.
“A lot” could also be used for the above scenario. But for more neutral situations where having a lot to do is not a negative thing, I would recommend “a lot” over “so much”.
“So much to do” is not overly rude, but I’d still personally avoid it in certain situations where I’m being extra polite or don’t want to sound like I’m complaining. For example if I were asking for a favor from a teacher: “I have a lot to do this evening, could I please have an extra day to finish this assignment?”
