@John31 Both are prepositions, but 'of' is possessive while 'about' relates to something or someone. Ex) I read a book about dogs. Ex) She was a member of that band.

@John31 'About' doesn't sound natural here. You would probably hear something along the lines of '...to check students' understanding.' and if anything else is included it would use 'of'. Ex) Use the following questions to check your students' understanding of the given text/words/etc.
