They are mostly the same, although "vacation" implies going somewhere special. You could take two weeks off and simply stay home! But practically, these are used the same way. :)

@Yu129 Yeah, sorta, but a vacation usually doesn't involve work (unless you're working, but you take your entire family with you to the place you have to go... and it's a vacation for them). It's more like to go on holiday, to spend time relaxing somewhere, like people go to Disney World all the time on vacation, or to visit family.
I think you'd say 休み for what I mean. We call it vacation; Brits call it holiday.
