The phrase "who you are" asks a person to define traits or features that they feel characterize their identity.
The second question, "Who are you?" Can be interpreted two ways: first, as a direct and literal question (for example: "I am Katie.") and second, as in the prior statement asking "who you are."
It's difficult to understand what someone is asking when they use, "Who are you?" without first knowing the context of the conversation. Since, as I mentioned earlier, the question can be answered multiple ways. The second question can be interpreted in a range from a benign inquiry to (depending on the tone) an insult.

`Who are you?` is the stock phrase for asking a stranger to identify themselves. They would normally respond with their name. It's a little unusual; "Tell me about yourself" is maybe more common.
