A really basic but confusing question: Is it appropriate to say "Thanks a lot" , "Many thanks" and " Very helpful/informative" to express my gratitude? Not sure what would sound sarcasm .

“Thanks a lot” can come off as sarcastic sometimes but it depends on the situation and your relationship with the speaker! If in doubt, it’s probably safer to stick with “thanks heaps” and “thanks so much / thank you so much.”
“Many thanks” also works but is a little more formal!
“This is/that was very helpful/informative” is fine also :)

Thanks a lot or many thanks (the latter a bit more old fashioned) are fine
