If a Russian speaker hears either подпо́ле or подпо́лье, is it immediately and unambiguously clear which word it is? Is there always a clear difference in the way the two words are pronounced, both when spoken in isolation and when spoken in the context of a sentence?

in the cellar = в по́дполе (ударение на первое о). "В подпо́лье" тоже имеет основное значение in the cellar (underground - переносное).

The soft sign adds the yot (й) sound also. The actual minimal pairs (unstressed) I can think of: в пла́те - в пла́тье, в бало́не - в боло́нье.
I'm not sure whether the yot sound completely disappeared in fast speech. I think a trace of yot, even a little one, remains anyway.

Пример.
Консервированные овощи у меня в подполе хранятся.
В том клубе проходят подпольные бои.
Чаще подполье употребляется именно в значении illegal.
А подполе заменят на подвал.

There is no such a word подпóле. Only пóдполе.

подпОлье так и правльно.
слово пОдполе встречала в разговорной речи в деревне. правильнее будет пОдпол (т.е под полом). Подполе произнося с прелогом в ( пример консервы хранятся в подполе). Подпол в частном доме. В многоквартирном это подвал.
